Abstract: The substantial increase in the usage of plant-based natural health products (NHPs) is increasing day by day due to their minor risk potential compared to other chemical medications. The plants harvested for use as NHPs are mostly collected from natural habitat, which in many cases, leads to incorrect identification and nomenclature. Furthermore, during the manufacturing processes of these plant-based NHPs, there is a risk of adulteration, both intentional and unintentional. In order to minimize these risks, DNA-based molecular methods such as qPCR and dPCR are widely used for proper qualitative and quantitative analysis. In this thesis study, I developed an effective detection and quantification methods for such adulterated NHPs with Actaea racemosa (Black Cohosh) as a model. In the first chapter, I valided and optimized a qPCR procedure for the identification of Actaea racemosa NHPs using a species-specific hydrolysis probe. In the second chapter, I developed an efficient quantification method for identifying and quantifying Actaea racemosa NHPs using qPCR and dPCR techniques.
