Bisphenols A and F induce Apoptosis in Bovine Granulosa Cells Via the Intrinsic Apoptotic Pathway

With the gradual decline in global fertility rates, there is a need to identify potential contributing factors and determine possible solutions. Endocrine disrupting compounds (EDCs), such as bisphenol A (BPA), are environmental toxicants that are known to negatively impact reproductive functions. As such, BPA has slowly been replaced with analogs, including bisphenol F (BPF) and bisphenol S (BPS), despite limited knowledge available regarding their potential to impact health. The following thesis investigates the effects of bisphenols A, S and F in bovine granulosa cells, with the goal of determining how they may impact oocyte competence and fertility. Determining the expression of key players in the intrinsic apoptotic pathway also allows us to establish potential mechanisms of action. The results of this thesis support that BPA and BPF reduce cell viability through the induction of apoptosis via the intrinsic pathway of apoptosis, while suggesting that BPS acts through a different mechanism.
